Yeah but from what i've heard the regeneration of outer hair cells is not sufficient enough to completely erase hearing loss. It regenerates only a few hair cells, the main purpose of this drug remains to regrow synapses.

Otonomy OTO-413 — Treatment of Hidden Hearing Loss

Oops, I forgot that spiral ganglion neurons remain active for decades in humans, so the synapse regeneration is possible whatever the date the noise trauma occured.

Otonomy OTO-413 — Treatment of Hidden Hearing Loss

They're talking about improvement when the subject gets injected OTO-413 the day following the noise trauma. So it doesn't work for people who got noise trauma years ago?

    Frequency Therapeutics — FX-345

If FX-345 could reach 20-4 kHz, then it's great, but aren't the hair cells responsible for speech frequencies below 1 kHz-2kHz?

Frequency Therapeutics — Hearing Loss Regeneration

I do think everyone experienced improvements. Severe hearing loss sufferers had great response because they have more inner ear hair cell death, so the hearing improvement can be easily seen unlike in other types of hearing loss.

    Frequency Therapeutics — FX-345

So FX-345 is exactly the same drug as FX-322 but goes deeper into the cochlea, am I right?
  7. Trevor_phillips

Frequency Therapeutics — Hearing Loss Regeneration

The new drug could be a FX-322 with improved delivery method, so it can reach the cochlea more profoundly or a completely new drug with new compounds, new delivery method (I hope nanoparticles) and which can reach the cochlea from the apex to the base.

Otonomy OTO-313 — Treatment of Tinnitus

I think that resolving tinnitus by restoring hearing function or indirectly by OTO-313 won't lead to silence for me. In my case I have spontaneous acoustic otoemissions in my healthy ear. In the other ear I have tinnitus and the sound difference is quite significant. Spontaneous acoustic...

Frequency Therapeutics — Hearing Loss Regeneration

Not sure but I do think that Otonomy gel can hit the cochlea from the apex to the base. I'm saying that because OTO-413 has recently proved clinically meaningful improvements in word recognition in noise. So basing on the fact that speech frequencies are almost at the base of the cochlea, I...

Otonomy OTO-6XX — Hair Cell Regeneration

I think it varies according to the company behind the product. I'm sure that they'll advance first OTO-413 and OTO-313 to get them to the market and after moving OTO-6XX to clinical trials. They'll have more data to successfully develop OTO-6XX.

    Pipeline Therapeutics

When will the trial results be released?

Frequency Therapeutics — Hearing Loss Regeneration

Why wouldn't FX-322 work on people with mild hearing loss? I don't get it because we know that the hair cell architecture is more intact in this degree. Therefore there should be more support cells and FX-322 would work at its best.

Frequency Therapeutics — Hearing Loss Regeneration

If Frequency Therapeutics doesn't work on reformulation and deeper penetration, I don't see how it could be successful.

Frequency Therapeutics — Hearing Loss Regeneration

Absolutely nothing is known about OTO-6XX. They did say that they are going to claim any information when they'll have an update to provide about the current pre-clinical tests. I won't be surprised if this is after OTO-413 results in mid-2022. I think that OTO-6XX will move to clinical trials...

Frequency Therapeutics — Hearing Loss Regeneration

If I take the example of a teacher explaining a lesson to his students in a large auditorium, assuming that all students have excellent hearing, the teacher will still need a microphone to properly interact with the whole room. This is what I want to explain. A minimum amount of amplification...

Frequency Therapeutics — Hearing Loss Regeneration

I do agree. This is why I'm looking to synaptogenesis drug like OTO-413 or PIPE-505 which have verified proofs. The recent results showed that FX-322 is a scam. The 10 dB improvements at 8 kHz and 6 kHz aren't reliable, audiograms can vary a lot. Furthermore, like @GlennS said above, it's...

Frequency Therapeutics — Hearing Loss Regeneration

I think this is because the untreated hair cells continued to accumulate continuous damage (if his tinnitus is different than the previous one) or the regrown hair cells are just not sustainable, and if this is the case, FX-322 is useless. Everybody wants regrown hair cells that can last for life.

    Sound Pharmaceuticals (SPI-5557 & SPI-1005)

Does SPI-1005 treat visual snow caused by previous cochlear damage? I read that it reduces the neuro-inflammation, then it could reduce some of the central hyperexcitability? Visual snow bothers me more than tinnitus, especially in the dark. Thanks.

Frequency Therapeutics — Hearing Loss Regeneration

This is the reason why I think it is better to get FX-322 first (assuming it comes to the market) and afterwards a synaptogenesis drug (OTO-413 or PIPE 505), because if the hair cells aren't able to form proper synaptic connections to the SGN, then a synaptogenesis drug will be needed. I...

Frequency Therapeutics — Hearing Loss Regeneration

I have noxacusis too, but I definitely think that cochlear synaptogenesis drugs (OTO-413, PIPE-505) will provide more relief than FX-322, basing on the assumption that OHC Type 2 synapses are damaged/destroyed in noxacusis. If FX-322 does mostly regrow IHCs in vivo, then it will have a minor...

Frequency Therapeutics — Hearing Loss Regeneration

I don't think that's the case for everyone. It's pretty common, especially for young people with good habits to have 0 dB loss or even +5 dB gain on every frequencies. It was my case before noise damaged my hearing which drove me to noxacusis and hearing loss at a certain degree. I would do...

    Pipeline Therapeutics

Considering PIPE-505 regrows some OHCs (unlike OTO-413) and mostly synapses: If PIPE-505 shows results as good as OTO-413 and audiogram improvements (if OHCs do regrow), is it better to get it rather than OTO-413?

Otonomy OTO-6XX — Hair Cell Regeneration

If OTO-6XX gel is the same as OTO-413 and consequently target a large part of the cochlea, is there any chance for this drug to outperform FX-322?

Frequency Therapeutics — Hearing Loss Regeneration

I have one question: Since the highest concentration of FX-322 is in the high frequencies, does it mean that the more high frequencies we can hear (for example up around to 17 kHz and nothing above), the better the results will be? Or there will be less place to PCA?

Frequency Therapeutics — Hearing Loss Regeneration

After you receive FX-322 injections at the ENT (if we assume FX-322 is successful in the clinical trials and becomes available to all of us): Can taking food supplements such as Ginkgo biloba, Acetyl-L-Carnitine, and Magnesium enhance cellular/synaptic growth and help in regeneration quality?

Therefore, acoustic otoemissions should be restored? I recently took one knowing about the fact that they are more noticeable in high frequency areas.

Frequency Therapeutics — Hearing Loss Regeneration

Does anyone think that regular head movements and listening to various sounds can help FX-322 to penetrate further in the cochlea and target more frequencies?

Frequency Therapeutics — Hearing Loss Regeneration

Do you think FX-322 could not only help hearing, but also vision? I got visual disorders (Visual Snow Syndrome) after getting hearing loss from noise trauma. I want to know if FX-322 restores my hearing, would my brain consequently readjust my vision? Thank you.

Will FX-322 be available first in the US and then later on in Europe through Astellas? Or will it be available at the same time in the US and EU?

Frequency Therapeutics — Hearing Loss Regeneration

My outer hair cells are probably damaged because of noise. Consequently, I have constantly "swinging hair cells" and it produces tinnitus for me on the frequency in which the problem is. That's a rare cause of tinnitus but i have it. Do you think that FX-322 could potentially help me? I don't...
