Having taken an antidepressant for years I have come to the conclusion that everything in my body has been affected by it and my nervous system in particular. The side effects are many, with more to come I feel.

Taking something else that targets the brain fills me with dread a little. I can't see any drug being without side effects.

That is correct, we were due to interview them today. I had a preliminary call with Justin and Rick last week, and they were really nice and keen to collaborate with us. However, unfortunately, we've had to postpone the recording. This is entirely my fault. I am currently staying with my mom, for work and family reasons, which I had not originally planned. This means I don't have access to a quiet space for recording. Being a volunteer project, the Tinnitus Talk Podcast cannot afford to rent a professional recording studio, we have to record from home. But we care a lot about audio quality, which is why we felt it better to postpone. We will reschedule for January, or February at the latest. My sincere apologies for the delay, but we hope to make it worth your while! :)

PLEEEASE ask him about chronic tinnitus and if this would be useful for that and what the evidence was AND if it is something you'd need to take daily in perpetuity. Puh Leeeez!

Just read through this thread, how encouraging! I'd also be intrigued to know whether they feel this treatment would help alleviate hyperacusis as well as tinnitus - hoping this question can be addressed on the Tinnitus Talk Podcast!

      @Justin De Moss

What do you think about this?

      20191218_102608.jpg

I am thinking that our brains are always wanting to ring, because that's what sound is, your auditory cortex is the sound generator, it is not receiving the sound signal. Silence in this case would be a full signal from the auditory nerve and sound would be the absence of the signal, like the opposite of a speaker wire. This would explain why noise damage could immediately induce tinnitus while walking into a silent room would not. It would also explain why older people that lose hair cells would not have tinnitus, because their nerve is still functioning. It would also explain how your pill could treat tinnitus, because it would allow the nerve to heal and begin sending the silencing signal again.

The question is why do antioxidants allow nerves to heal and is this only acutely? The Hough pill is a very powerful free radical scavenger and antioxidant, not a growth factor.

From my understanding, Anti-Oxidants prevent the nerves from dying by attacking free radicals before they can do damage. Which is why I also asked if this could be used beyond 2-3 day window from damage onset, just not sold on that yet. I hope I'm wrong.
I'm not really worried about Anti-Oxidants. Probably one of the safer approaches, of course too much of a thing is bad for us, so perhaps the amount they need to get this formula to work, is in excess of what our bodies can handle. As with everything else, only time will tell. Hopefully we'll know sooner rather than later.

They finished Phase 1 in 2015. It could have been in Phase 3 or in the market by now.

I wish they got enough funding back in 2015 so we could see it in Phase 3 or released in markets by now.

Maybe the PRP + Dexamethasone injections would have helped you more if you had gotten them within 24 hours of your tinnitus onset.

I truly believe that. Not even PRP more so than dexamethasone. Yeah, you're right. I went to my doctor within 48 hours and he gave me oral steroids.

Considering how chicken ENT's are to administers IT injections the existence of that pill probably would have been more plausible only if.

Hey there, just a kind request to hold off on sending your questions directly to Hough at this point, please just post them here and wait for the podcast episode. That's more efficient for Hough, and the answers will reach a much wider audience than just the people reading this thread. Thanks!

Ha ha, just looked up the thread to see the same post.

I noticed when looking at their site that at least half of their research team are from China. I wonder why that is? Not that it matters, just interested.

If the pill is just a high potency anti oxidant and free radical scavenger, shouldn't things like Astaxanthin and high dose vitamin C at least partially have the same response? There are no actual nerve growth factors in the pill.

"Surprisingly, the observation that HPN-07 treatment initiated within four hours after noise exposure and continued for 2 days has long-lasting effects, some of which only become evident 21–180 days later, is not readily explained from our evaluations but could be interpreted to suggest that HPN-07 may have the capacity to initiate repair mechanisms or a regenerative response among one or more components of the cellular architecture of the cochlea."
"In summary, the physiological and histological findings presented here indicate that acute post-trauma intervention with HPN-07 as a single therapeutic agent promotes both acute and chronic recovery of hearing function."

Looks like they are guessing that it could treat chronic tinnitus and hearing loss and they didn't actually test it in chronic models. I could be wrong.

Update: Looks like I was wrong. See below.

Gather 'round little children, have you a seat there on Papaw's lap:

FIG. 1 shows that HPN-07 induced a neuritogenic response in murine spiral ganglion (SG) tissue explants in vitro. HPN-07 induced marked neurite outgrowth relative to untreated controls cultured in the same organotypic culture medium (NC, normal control). Both the number of neurites radiating out from each SG explant and the corresponding length of each of these extensions increased in the presence of HPN-07.

FIG. 30 shows NHPN-1010 treatment (HPN-07 plus NAC) resulted in recovery of ABR Wave I amplitudes in established chronic hearing loss model.

Thanks, Papaw. This surely means the currently available anti oxidants have benefit past the acute stage then, too, right? At least partial benefits since HPN-07 is stronger.

Not necessarily. We can't assume that just because it is an anti-oxidant that it is only an anti-oxidant. Take curcumin for example. It's an anti-oxidant but it is also a verified potent notch1 inhibitor.
