I've been in S. Korea for the last 2 weeks getting PRP injected into my ears at Dr. Shim's Solsaem clinic in Seoul. I will answer your questions as well as periodically update my status. I've got between 2 and 3 months for the full effects to manifest.

Here's how I think Minbo Shim's therapy works. He didn't explain this to me, I put all of this together myself.

First off, this is how hair cell regeneration technology works.

A class of chemicals called notch inhibitors or gamma secretase inhibitors makes contact with the surface of the stem cell. The stem cells in our inner ear are classified as LGR5+ (positive), meaning that it expresses a gene called Leucine-rich repeat-containing G-protein coupled receptor 5.
      https://en.wikipedia.org/wiki/LGR5

Inside the cell there is a protein called Beta-Catenin. This is the finger that throws the "switch" that makes LGR5+ stem cells proliferate, which means divide and regenerate into mature cells, in this case, hair cells in the cochlea. In order for this to occur, enough Beta-Catenin has to enter into the nucleus of the cell.

But, the reason our hair cells aren't regenerating on their own is because once Beta-Catenin is created inside the cell, there is a group of proteins that form something called the Beta-Catenin destruction complex, this destroys the Beta-Catenin before it can enter into the nucleus in enough quantity to activate the proliferation process.

      The β-catenin destruction complex.
      https://www.ncbi.nlm.nih.gov/pubmed/23169527

One way to stop the Beta-Catenin from being destroyed is by activating a cellular signalling pathway called WNT (pronounced wint).

Generation of hair cells in neonatal mice by β-catenin overexpression in Lgr5-positive cochlear progenitors.
      https://www.ncbi.nlm.nih.gov/pubmed/23918377

This is how drugs like FX-322 work to "wake up" dormant stem cells in the inner ear.

There is a class of proteins involved with many functions in our bodies called growth factors.
      growth-factor-description.png

One of these growth factors is called insulin-like growth factor 1. IGF-1.

IGF-1 has been shown in multiple human clinical trials to improve hearing recovery.

Insulin-like growth factor 1: A novel treatment for the protection or regeneration of cochlear hair cells.

      https://www.ncbi.nlm.nih.gov/pubmed/25937136

Actually I never noticed this part before but they verified that it causes cochlear supporting cells to proliferate in cochlear explants.

"The mechanisms of IGF1-induced maintenance of hair cell number have been investigated using a cochlear explant culture system, which demonstrated that IGF1 acts on supporting cells, leading to the inhibition of hair cell apoptosis and the proliferation of supporting cells."

Another study, showed that IGF-1 increases cellular Beta-Catenin levels.

Insulin and IGF-1 stimulate the β-catenin pathway through two signalling cascades involving GSK-3β inhibition and Ras activation

      https://www.nature.com/articles/1204064

"IGF-1 increased the cytoplasmic levels of β-catenin."

IGF-1 is produced in our bodies, in part in blood platelets.

There is not any blood in our inner ear fluid. Inner ear fluid is different from blood.

      https://en.wikipedia.org/wiki/Endolymph

You can create a platelet rich composition of blood by putting drawn blood in a centrifuge which separates the platelets into something called platelet rich plasma, PRP.
PRP contains growth factors, including IGF-1.

The growth factors and other cytokines present in PRP include:

      https://en.wikipedia.org/wiki/Platelet-rich_plasma

In your middle ear there is a semi-permeable membrane called the round window.

      the-middle-ear-round-window.png
      https://en.wikipedia.org/wiki/Round_window

So that's how you can deliver medicine to the cochlea, by filling up the middle ear, via eardrum injection, with a viscous solution containing X substance, it rests in the middle ear and the substance, if molecularly small enough, will diffuse through the round window membrane. That's why the various presentations produced by FrequencyTX, as well as their homepage emphasize that their products are "small molecule drugs", because this is exactly how FX-322 is delivered to the cochlea.
      frequency-therapeutics-fx-322.png

Other neurotrophic compounds are also being investigated to repair lost cochlear synapses by diffusion through the round window membrane.

Round-window delivery of neurotrophin 3 regenerates cochlear synapses after acoustic overexposure

      https://www.researchgate.net/public...cochlear_synapses_after_acoustic_overexposure

So that's my theory, that IGF-1 works just like FX-322 to disable the Beta-Catenin destruction complex and cause the stem cells to proliferate into functioning hair cells except that IGF-1 is already produced in our bodies and needs to be manually introduced into the cochlea. Also PRP may be much safer than FX-322 because it is good for healing holes to the eardrum that would be caused by the injection.

Topical use of autologous platelet rich plasma in myringoplasty.

      https://www.ncbi.nlm.nih.gov/pubmed/25794691

"Topical autologous PRP application during myringoplasty is safe and highly efficient and successful with no reported complication"

      https://www.ncbi.nlm.nih.gov/pubmed/25794691

Now watch the nay-sayers purposefully push this post up the page with extremely long-winded ad-hominem attacks on me and Minbo Shim.

As of now, my tinnitus in my right ear has gone from two tones, a high hiss around 12-14 kHz to almost gone. Also in my right ear was a distinct lower tone at about 6.5 kHz and that has diminished to basically nothing and I have to plug my ear to hear it.

He spent the last few days focusing on my left ear which was way more jacked up. I have the same two tones in my left as my right, and I had suspected a lower tone for sometime and it was very clear in the audio booth. They determined that tone was about 250 Hz. Yesterday in the audio booth I noticed that tone was gone. Now the hiss, which was in the middle of my head has moved primarily to the left side.

My left side was the side facing my guitar amp when my acoustic trauma occurred. I am already very satisfied with the improvemnt in my right side, now I'm just waiting/hoping for my left to catch up.

Also here is an insight I should have known. I had taken a hearing test with a phone app and it made me calibrate the headphones before the test. Obviously, you cannot do that to check your own hearing loss duh. The professional audiogram revealed much more meaningful information about my hearing loss and tinnitus.

Oh yeah, the first day he also diagnosed me with rhinitis and gave me surgery for that as well. That was fun.

@JohnAdams Damn, a lot of people talked, but you actually muscled up and did it. Much respect for taking a huge risk in traveling half way around the world and having an experimental treatment performed. Glad that the procedure went well for you man, looking forward to your future updates regarding progress. I'd love to get some of my hearing back, the connection between hearing loss and degenerative neurological conditions is somewhat worrisome to me. Lowered tinnitus is just icing on the cake.

I had tinnitus for as long as I can remember. Got bad later.
I cannot even fully process how happy this makes me. After two years of trying to keep the discussion and investigation of this doctor alive, I am overjoyed that all the time I put into this paid off and somebody was able to be treated. Thank you so much for the time you put in researching the science based on his method, and especially thank you for your courage for actually going through it. I know full well that the treatment wasn't a fun experience, and it is great that it ultimately paid off. The result of your journey is going to help a lot of people.

I would seriously hug you right now. You're awesome.

      Hi @JohnAdams,
      Thanks for the update.
I live not too far away, may pay them a visit and try that.

      How much did you pay?

Since when did you have tinnitus and what noise did it cause?

No way tinnitus free. My hearing and tinnitus in my right ear has improved significantly and there have been minor gains in my left ear. The whole thing takes 2-3 months to fully realize the results. I want to reiterate that my improvemnt in my right ear is both an improvemnt in hearing and a decrease in tinnitus.

Platelet-Rich Plasma: A Promising Product for Treatment of Peripheral Nerve Regeneration After Nerve Injury -- The following is the introduction to this research article...

Nerve regeneration includes regrowth of injured axons as well as myelination, restoration of synaptic connections and recovery of physiological functions. Platelet-rich plasma (PRP) is prepared from the patient's own blood and contains growth factors that influence wound healing and used in various surgical fields including oral and maxillofacial surgery. When platelets are activated either ex vivo or in vivo, growth factors and proteins were released from platelets' alpha granules. Recent studies proved that PRP could promote regeneration of injured peripheral nerve.

I'm the first on this board and the first America. My 8 kHz audiogram shows a -15 dB dip at 6.5 kHz in both ears and my HF audiogram is kinda scary and it's the HF tinnitus that really bothers me.

Well this is new. I spoke to him on Skype back in October. This is how he replied to me. I had some doubts as before even understanding my scenario he said that I am not ready to be treated yet whatever the hell that means.

How on earth can this person be trusted? Clearly we need more reviews or feedback. He quoted me back then $7,000. I was even willing to go but his replies put me off...

Below is the copy of the transcript:

      Amit, 12:08
      Where are you based

      Amit, 12:08
Why your address is not on the website

      Amit, 12:08
      Why are you in russia?

      Amit, 12:09
What does treatment involve can you tell?

      Amit, 12:09
      What is cost of treatment?

      Minbo, 14:15
So many doubts. I understand .But you have to know that I have no duty of treating you.

      Amit, 14:16
Are you able to answer these questions i have seen cost on another website but i dont understand you mention Korean standard time and your patent is registered in russia

      Amit, 14:17
I have no problem to pay but i wish to know the answers to my questions

      Amit, 14:17
As you know no cure exists for tinnitus

      Minbo, 14:18
I will give you one last chance to make me want help you. If you fail I will block you.Again I dont have to answer those qs.

      Amit, 14:19
Ok can you explain what i need to do i have noise induced tinnitus last 2 years got worse as was exposed to loud music

      Amit, 14:19
I dont usually go to clubs never infact

      Amit, 14:20
In total ive had for 30 years

      Amit, 14:21
So if you can help me please tell me what to do.

      Minbo, 14:25
You are not prepared to be treated yet.

      Amit, 14:25
      Im not prepared means sorry?

      Minbo, 14:25
If you are ready, mail to solsamclinic@gmail.com

      Amit, 14:26
I am ready i just wanted to know more information

      Minbo, 14:26
      No. You are not ready.

      Amit, 14:27
      So is this another clinic

      Amit, 14:29
If your able to treat me why do you say i am not ready. I have damaged inner hair cells if they can be fixed im more then ready.

      Amit, 14:32
Do you wish to help me or not?
